Specific killing effect of CIK induced by CEA-rV-bearing DC on CEA positive primary tumor cells

Abstract: Objective: To investigate the specific killing effect of dendritic cells (DC)-induced killer cells (CIK) carrying CEA recombinant vaccinia virus (CEA-rV) antigen on CEA-positive primary tumor cells. Methods: Cord blood mononuclear cells (UBMC), DC, CIK, DC + CIK, CEA-rV + DC + CIK and CEA-rV + DC + UNBC were prepared from cord blood. Freshly resected esophageal cancer tissues and lung cancer tissues were taken to prepare primary cultured CEA positive tumor cells as target cells. MTT method was used to measure the killing activity of 5 groups of effector cells to two kinds of target cells. Results: 1) Two primary cultured target cell lines were identified with CEA rabbit anti-human monoclonal antibodies. 2) the molecular phenotype of DC was determined by flow cytometry, and it was confirmed to be DC. 3) CIK CD3 + CD56 + double positive cells were determined by flow cytometry 75.4%. 4)DC + CIK compared with CIK, the two kinds of CEA positive tumor cell killing rate difference was not statistically significant, P>0.05. 5) the killing rate of CEA-rV + DC + CIK on two kinds of CEA positive tumor cells (78.80%) was significantly higher than that of DC + CIK(52.94%),P<0.05. Conclusion: CEA-rV + DC can significantly improve the killing activity of CIK cells on primary cultured CEA positive tumor cells, while DC alone can not improve the killing activity of CIK, indicating that CEA-rV plays an important role in improving the specific killing effect of CIK on CEA positive tumor cells.
