Comparison and application of biological characteristics of CIK cells in healthy people and cancer patients

Abstract:Objective To compare the in vitro expansion rate, cell phenotype and killing activity of CIK cells derived from peripheral blood of healthy people and cancer patients, and to observe the preventive and therapeutic effects of healthy human CIK cells on transplanted tumors in nude mice. Methods 10 peripheral blood mononuclear cells from healthy people and tumor patients were induced into CIK cells, and the amplification speed of the two cells was compared by direct live cell counting method, the cell phenotype of the two cells was compared by flow cytometry, and the killing activity of the two cells on K562 and LOVO cell lines was compared by MTT method. Thirty nude mice were divided into three groups, prevention group: first tail vein injection of CIK cells, back subcutaneous inoculation of A549 cells. Treatment group: nude mice were inoculated with A549 cells on the first day. On the second day, according to the different injection schemes, they were divided into three groups: local injection of CIK cells (local injection group), tail vein injection of CIK cells (tail vein injection group), local and tail vein injection of CIK cells (combined treatment group). Control group: A549 cells were inoculated subcutaneously on the back, and normal saline was injected for 5 days. After 4 weeks, the tumor size of nude mice was measured, weighed, the tumor volume and tumor inhibition rate were calculated, and pathological examination was performed. Results The expansion rate of CIK cells in healthy people was significantly higher than that of CIK cells in tumor patients (P<0.05).
