Experimental study on the inhibition of nerve cell apoptosis in the brain tissue of HIBD rats by acupuncture

Abstract:Objective To investigate the protective effect and mechanism of acupuncture on neuronal apoptosis in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods (1) 85 7-day-old clean SD rats were selected. The rats were ligated with the left common carotid artery and recovered for 2 h. They were placed in a transparent airtight container, incorporated into 37 ℃ constant temperature water and introduced hypoxic gas at a speed of 1 L/min. After 2.5 h, the animals were taken out. The survivors continued to conduct behavior tests. 72 successful models were randomly divided into 3 groups, group A was HIBD acupuncture group I, group B was HIBD acupuncture group II, group C was HIBD control group, and group D was sham operation group, with 24 rats in each group. (2) Young mice Baihui, affected temporal I, Neiguan, Quchi, Zusanli and Yongquan were selected. Group A started acupuncture 24 h after modeling, only limb acupuncture points were injected for the first 7 days, and head acupuncture points were added on the 8th day. Acupuncture was started on the 8th day after modeling in group B, and head and body needles were synchronized. Acupuncture was performed once/d, group A was treated with continuous acupuncture for 20 d, group B was treated with continuous acupuncture for 13 d. The intervention measures in sham operation group were the same as those in group A, and the materials were killed on the 21st day after HIBD. The apoptosis of neurons in the frontal lobe and hippocampus of HIBD rats was measured by TUNEL method, and the number of apoptotic neurons was calculated by image analyzer under light microscope. Results There were significant differences in the number of apoptotic neurons in the left hippocampal CA1 region and the left frontal cortex among the four groups (P<0.05).
