Study on the killing effect of specific CIK cells induced by CEA-rv on Lovo cell line

Abstract:Objective To investigate the effect of carcinoembryonic antigen recombinant vaccinia vaccine (CEA-rv) loaded with cord blood-derived dendritic cells (DCs) on the cytotoxic activity of colon cancer cell line Lovo induced by CIK cells. Methods The prepared CEA-rv were used to prepare DCs and CIK cells from cord blood mononuclear cells (CBMC), respectively. The CEA-rv-DC-CIK cells were induced by co-culture with CEA-rv loaded DCs and CIK cells. The DC immunophenotype was detected by flow cytometry, and the cytotoxicity of CBMC, CIK, Ag-CIK, Ag-DC-CIK and CEA-rv-DC-CIK against Lovo tumor cell line was detected by MTT assay. Results CD86 and CD40 were highly expressed in cord blood DCs, and CD83 and CD80 were moderately expressed. The killing ability of Lovo cells (62%) in the experimental group (CEA-rv-DC-CIK) was significantly higher than that in the control group (P<0.01). Conclusion CEA-rv loaded DCs can enhance the cytotoxicity of CIK cells to Lovo cells, which provides a new idea for the immunotherapy of CEA positive tumors.
