Clinical efficacy of umbilical cord blood mesenchymal stem cells in the treatment of severe systemic lupus erythematosus

Abstract:Objective To investigate the therapeutic effect of umbilical cord blood mesenchymal stem cells on severe systemic lupus erythematosus (SLE). Methods Mononuclear cells (MSCs) were isolated from umbilical cord blood (UCB) of full-term pregnant women and cultured to prepare mesenchymal stem cells (MSCs). MSCs culture to the third generation, sampling UCB-MSCs, using flow cytometry for characterization showed that: UCB-MSCs high expression of CD29,CD90; no expression of CD34 and CD45. Collect well-grown third-generation UCBMSCs. (5.0~6.0) * 107 cells are added into 100ml of normal saline containing 1% human albumin for intravenous drip. Once a day, 6 ~ 10d, a course of treatment; The second course of treatment will be used after 3~6 months. Results Through UCB-MSCs infusion, the symptoms of 2 patients with severe systemic lupus erythematosus who were ineffective in the treatment of the existing methods were completely restored. The first case of SLE was accompanied by severe lupus kidney, lupus brain for 10 years, edema of both lower limbs, skin gangrene, ulcer deep to bone surface, severe pain and sleeplessness for 1 year. The doctor mobilized her to amputate and save her life. UCB-MSCs 10 days after treatment, the condition improved significantly, and the edema of both lower limbs almost completely subsided. After 6 months of treatment, 10 of the 11 gangrenous and ulcerated sores were basically healed. After 10 months, the patient's gangrene, ulcer sores all healed, can move freely, into normal life.
