Effects of Acupuncture and Nerve Growth Factor on Intelligence and Apoptosis Genes in Hippocampus of Young Rats with Cerebral Palsy

Abstract:Objective: To observe the effects of acupuncture and nerve growth factor (NGF) on the intelligence recovery and the expression of BCL-2, BAX and Casp3 mRNA in the hippocampus of cerebral palsy rats. Methods: The model of cerebral palsy in young rats was prepared by the method of ischemia and hypoxia (HIBD), and the young rats were weighed 7 days after birth, divided into experimental groups and modeled. The experiment was divided into five groups: normal group, sham operation group, model group, simple acupuncture group and acupuncture NGF group. Postoperative 3~15 d according to the plan to give the corresponding intervention. The mRNA expression of BCL-2, BAX and Casp3 gene in hippocampus was detected by fluorescence quantitative PCR(SYBRGreen I dye method) at 18 days after operation, and the apoptosis of cerebral cortical nerve cells was detected by TUNEL method. Results: Compared with the model group, the value of anti-apoptotic BCL-2 gene mRNA expression in the brain cells of the simple acupuncture group and the NGF group was higher; the value of BAX and Casp3 gene mRNA expression was lower; the apoptosis index in the brain cortex was lower, and the apoptosis index in the acupuncture NGF group was lower than that in the simple acupuncture group. Conclusion: Acupuncture has obvious therapeutic effect on HIBD rats in terms of anti-apoptosis of brain nerve cells, and the effect is better after adding NGF.
