Study on the killing effect of CIK on CEA positive tumor cells induced by CEA-rV loading DC

Abstract: Objective To study the killing activity of CEA-positive tumor cell lines by inducing CEA antigen-specific cytokine-induced killer cells (DCs) after CEA-rV loading of cord blood-derived dendritic cells (DCs). Methods Umbilical cord blood mononuclear cells were isolated with lymphocyte separation solution, DC, CIK cells and CEA-rV loaded DC were induced respectively, and then mixed with CIK cells to induce CEA-specific cytotoxic T lymphocytes. CEA-positive and CEA-negative tumor cell lines were killed by the above different types of cells, respectively. Results The killing activity of specific CIK induced by CEA-rV on CEA positive tumor cells was 58.2% for Lovo and 62.4% for A549, which was higher than that of DC-CIK group on CEA positive tumor cells was 44.8% and 50.2% for A549, and the difference was statistically significant (P<0.05). The killing activity of specific CIK induced by CEA-rV to CEA negative tumor cells was 79.7% in K562 group and 70.1% in Bel7402 group, 78.7% in K562 group and 67.8% in Bel7402 group, and there was no significant difference between the two groups (P>0.05).
