Clinical application of cord blood CD3AK cells in the treatment of malignant tumors

Objective: Objective: To study the anti-tumor effect of CD3AK cells prepared by umbilical cord blood mononuclear cells, and to explore the immune indicators of the short-term efficacy of tumor biotherapy. Methods: Cord blood mononuclear cells were isolated, and LAK and CD3AK cells were induced by IL-2 and IL-2 + CD3Ab, respectively, and the number of expansion and the killing activity of K562 cells were measured. The changes of absolute T lymphocyte subsets and NK killing activity of peripheral blood mononuclear cells (PBMC) were measured before and after treatment with CD3AK cells. Results: Umbilical cord blood LAK cells and cord blood CD3AK cells had the highest amplification times at 11 days after culture, which were 18 times and 24 times respectively before culture, and their killing activity against K562 was 2.6 times and 3.2 times respectively before culture. After a course of CD3AK cell infusion, the absolute count of peripheral blood T lymphocyte subsets in tumor patients increased significantly: total T increased by 66.0 and Th increased by 68.0, ts increased by 58.0; The NK killing activity of PBMC increased from 63.0 to 81.0, with an average increase of 28.0. Conclusion: 1) Umbilical cord blood mononuclear cells are good precursors of LAK cells and CD3AK cells. 2) The number and killing ability of LAK and CD3AK cells reached the peak at 11 days of culture, and the number and killing activity of CD3AK cells were significantly better than LAK cells. 3)CD3AK cell infusion can significantly improve the absolute count of peripheral blood T lymphocyte subsets and NK activity of PBMC in tumor patients. 4) The peripheral blood T lymphocyte count and NK activity of PBMC in tumor patients are expected to be a short-term efficacy parameter of tumor biotherapy.
