Preparation, biological activity and clinical application of CD3AK cells from umbilical cord blood

Abstract: Objective To study the anti-tumor effect of CD3 AK cells prepared from umbilical cord blood mononuclear cells, and to explore the immune indicators of the short-term efficacy of tumor biotherapy. Methods Cord blood mononuclear cells were isolated, and LAK and CD3AK cells were induced by IL 2 and IL 2 + CD3 AB, respectively, and the number of expansion and killing activity against K5 6 2 cells were measured; NK killing activity of peripheral blood mononuclear cells (PBMC) in tumor patients before and after treatment with CD3 AK cells was measured. Results Umbilical cord blood LAK cells and cord blood CD3 AK cells were the highest expansion times on the 11th day after culture, which were 18 times and 24 times respectively before culture, and the killing activity to K5 6 2 was 2.6 times and 3.2 times respectively before culture. After a course of infusion of CD3 AK cells in tumor patients, the NK killing activity of PBMC increased from 62% to 82%, the average increase was 32%. Conclusion (1) Umbilical cord blood mononuclear cells are good precursors of LAK cells and CD3 AK cells. (2) The number and killing ability of LAK and CD3 AK cells reached the peak at the 11th day of culture, and the number and killing activity of CD3 AK cells were significantly better than that of LAK cells. (3)CD3 AK cells infusion can significantly improve the NK activity of PBMC in tumor patients. (4) Determination of NK activity of PBMC in tumor patients is expected to be a short-term therapeutic parameter of tumor biotherapy.
