Abstract:Objective To investigate the changes of cytokine secretion and cytotoxicity of cytokine-induced killer (CIK) cells before and after immunoediting. Methods Cord blood mononuclear cells were extracted by Ficoll density gradient method and induced into CIK cells, that is, CIK cells before immune editing. CIK cells and human lung cancer A549 cells were co-cultured in Transwell chambers, and CIK cells were isolated and purified, that is, CIK cells after immune editing. The levels of cytokine secretion and cytotoxic activity of CIK cells before and after immunoediting were detected by ELISA and MTT assay, respectively. Results After the co-culture of CIK cells and A549 cells, the contents of IFN-γ and TNF-α secreted by CIK cells decreased significantly (P<0.05), while the IL-2 decreased slightly. The cytotoxicity of CIK cells was significantly decreased after co-culture (P<0.05). Conclusion There is an immune editing effect between CIK cells and human lung cancer A549 cells, and the levels of cytokines secreted by CIK cells and the killing activity of CIK cells are decreased after editing.
