Study on the anti tumor immune effect of heat shock protein 70-peptide complex in lung cancer and its correlation with heat syndrome

Objective: Objective: To observe the anti-tumor immune effect of heat shock protein 70-polypeptide complex (HSP70-PC) on human lung cancer A- 549 cells in lung cancer patients with different syndromes, and to explore whether its anti-tumor effect is related to syndromes. Methods: ADP affinity chromatography, DEAE ion exchange chromatography, SDS-PAGE, Westem-blot, cell culture, MTT assay and other methods were used to isolate and purify HSP70-PC from human lung cancer tissues and distal normal lung tissues, and to observe the activation and proliferation effect of tumor-derived HSP70-PC on human peripheral blood mononuclear cells (PBMC) and the cytotoxic effect of sensitized lymphocytes on human lung cancer A- 549 cells. Results: ①Compared with the blank control group, the proliferation effect of lymphocytes in the PC group of heat syndrome and non-heat syndrome carcinoma was significantly increased; the sensitized lymphocytes had a strong killing effect on human lung cancer cells, and there was a significant difference compared with the blank control group. The effect of promoting lymphocyte proliferation and tumor cytotoxic killing in the PC group of heat-type cancer was significantly higher than that in the PC group of non-heat-type cancer. ③The HSP70-PC of the distal normal lung tissue of patients with lung cancer did not significantly stimulate the proliferation of lymphocytes, and the killing effect was not significantly increased. Conclusion: Human lung cancer tissue HSP70-PC can induce anti-tumor immune effect against human lung cancer cells, and its anti-tumor effect is likely to be related to heat syndrome.
