Dendritic cells derived from cord blood enhance the killing effect of CIK cells on lung cancer cell lines

Abstract: Objective To investigate the effect of human lung cancer cell line A549 tumor freeze-thaw antigen loaded cord blood-derived dendritic cells (DCs) mixed with CIK cells on the killing activity of A549 cells. Methods A549 cells in logarithmic growth phase were used to prepare tumor antigen (Ag), DCs and CIK cells were prepared from cord blood mononuclear cells (CBMC), and Ag-loaded DCs and CIK cells were co-cultured to induce Ag-DC-CIK cells. The immunophenotype of DC was detected by flow cytometry, and the killing activity of CBMC, CIK, Ag-CIK and Ag-DC-CIK on A549 tumor cell line was detected by MTT assay. Results DCs in cord blood had high expression of CD86 and CD40, moderate expression of CD83 and CD80, and low expression of CD11c. The killing ability of Ag-DC-CIK cells (62%) to A549 cells in the experimental group was significantly higher than that in the control group (P<0.01). Conclusion DCs with tumor antigen load can enhance the killing activity of CIK cells to target cells, which provides a new idea for the immunotherapy of DCs.
