Preventive and therapeutic effects of CEA recombinant vaccinia virus on experimental CEA positive hepatocellular carcinoma

Abstract: Objective: To investigate the preventive and therapeutic effects of CEA recombinant vaccinia virus (CEA-rV) on CEA-positive tumors in mice. Methods: ① Observation of preventive effect: Experimental mice were divided into 4 groups. Groups 1, 2 and 3 and 4 were subcutaneously inoculated with wild-type vaccinia virus (W-VV) and CEA-rV3 times respectively. After the last inoculation, groups 1 and 3 were subcutaneously injected with homologous mouse CEA-negative liver cancer cells (Hepa-CEA-), and groups 2 and 4 were inoculated with Hepa-CEA. Tumor volume was measured weekly for 6 weeks after tumor cell inoculation. Observation of therapeutic effect: groups 1 and 2 were injected with Hepa-CEA subcutaneously, groups 3 and 4 were inoculated with Hepa-CEA, groups 1 and 3 were inoculated with W-VV subcutaneously after 7 days, and groups 2 and 4 were inoculated with CEA-rV. Tumor volume was measured weekly for 6 weeks after tumor cell inoculation. Results: ①Preventive effect: There was no tumor formation in the mice of the inoculated CEA-rV group and the Hepa-CEA group (group 4) during the observation period, and the other mice had tumor formation under the skin, and the growth rate was similar. ② Therapeutic effect: The subcutaneous tumors of the mice in the inoculated Hepa-CEA and CEA-rV groups (group 4) grew slowly and the tumor mass was smaller, while the tumors of the other three groups grew rapidly and the tumor mass was larger. The mice in each group did not show any other adverse reactions during the whole experiment. Conclusion: CEA-rV has a good preventive and therapeutic effect on experimental CEA-positive tumors, and the preventive effect is better.
