Biological characteristics of CIK cells in healthy people and cancer patients

Abstract:Objective To compare the in vitro expansion rate, cell phenotype and killing activity of CIK(Cytokine-induced killer) cells between healthy people and cancer patients, in order to provide experimental data for the selection of effector cells in clinical application. Methods The peripheral blood of 10 healthy people and 10 tumor patients were centrifuged by density gradient centrifugation to obtain mononuclear cells (PBMC), and CIK cells were prepared with interferon-γ, CD3MAb, IL-2 and IL-1 as inducers. Results The amplification rate of CIK cells in the peripheral blood of healthy people and the killing activity of K562 and LOVO cell lines were significantly higher than those of CIK cells in the peripheral blood of tumor patients (P<0.01). Flow cytometry showed that the percentage of CD3 CD56 of CIK cells in the peripheral blood of healthy people was significantly higher than that of CIK cells in the peripheral blood of tumor patients (P<0.01). Conclusion Compared with cancer patients, CIK cells in peripheral blood of healthy people expanded faster in vitro, CD3 CD56 ratio was higher, and the killing activity was stronger. It provides an experimental basis for the clinical application of CIK cells for adoptive immunotherapy.
